From the comfort of League One, sure why not.

When we were in the Conference with just one up, Phil was one of the driving forces to get more promotion places.

Sure it might mean you fall out the "League" but increases your chances of coming back up again, so don't see it being a massive difference. There's a lot of side in the conference and League Two now that have been in both at some point. The established League sides should have a size advantage over the traditional non-league sides anyway.
